Why the Tissue Cultured Yangmei liners show the same leaf shape (serrated margin) as seedlings ?
It is normal phenomenal during plant tissue culture, called "Rejuvenation", especially when we do shoot tip culture and subculture many times. On the above pictures, you can see how small the in vitro shoots are, at the beginning, I cut even smaller!!!! The rejuvenation sometimes also happens when you do cuttings or grafting for many generations on some plants.

The genome size of Yangmei is 313-Mb. By whole-genome comparison between the female and male and association analysis with sequences of bulked and individual DNA samples from female and male, a 59-Kb region determining female was identified and located on distal end of pseudochromosome 8, which contains abundant transposable element and seven putative genes, four of them are related to sex floral development.
We follow the simple and effective protocol developed by Zhejiang University (https://patents.google.com/patent/CN108103219A/en). Both primer Mr-F1 and Mr-F2 are very effective. The female will show two bands of 470bp and 268bp (Mr-F1); or 426bp and 332 bp (Mr-F2). It clearly indicated our tissue cultured plants are female.
